Gradual decline of CD4⁺ T cells in cynomolgus macaques persistently infected with macaque-tropic HIV-1

Although macaque-tropic human immunodeficiency virus type 1 (HIV-1mt) strains with high genomic similarity to HIV-1 have been engineered, their potential to cause acquired immunodeficiency syndrome (AIDS)-like symptoms in monkeys is essential for advancing HIV research. In this study, we revealed that long-term HIV-1mt infection in cynomolgus macaques causes progressive CD4⁺ T cell depletion, indicating increased pathogenicity. In addition, we identified adaptive amino acid substitutions in viral proteins that enhance replication fitness in macaque cells. These findings indicate that extended in vivo adaptation increases the pathogenic potential of HIV-1mt and provides novel perspectives on viral evolution and adaptation to the host.
