抄録

アノイキスとは、細胞外基質との接着が不十分あるいは完全に喪失した際に惹起されるアポトーシスの一形態であり、その破綻はがんの浸潤や転移に直結する。本総説は、再生医療や組織工学を専門とする材料研究者のうち、細胞死の分子機構に関する系統的な知識を必要とする初学者に対し、アノイキスのシグナル伝達経路を俯瞰するための基礎的枠組みを提示する。培養皿から剥がした細胞が死ぬ現象を多くの研究者は経験しているが、それが偶発的な壊死ではなくプログラムされた細胞死であることを意識する機会は少ない。しかし、この現象の分子メカニズムを理解することは、細胞の生死を人為的に制御する材料設計への第一歩となる。本総説では、インテグリン–FAKを起点とした接着の感知から、ミトコンドリア外膜透過化を経てカスパーゼによる細胞解体に至るまでの情報伝達の流れを、各分子の役割に焦点を当てながら一気通貫で解説する。さらに、この基礎知識を基盤として、脱細胞化スキャフォールドや合成ハイドロゲル、マイクロ流路デバイスなどの具体的な材料技術にどのように応用されうるかを示し、最終的にはアノイキス制御による免疫応答調節という未踏のフロンティアへと読者を導く。本総説は、材料研究者が自らの扱う接着環境を分子レベルで解釈し、細胞の生存と死をデザインするための橋渡しとなることを意図している。
